Type I diabetic spontaneously hypertensive rats (SHR-1DM) have elevated blood pressure compared to SHR-1, whereas SHR-2, derived from SHR by selective backcrossing onto the normotensive Wistar-Kyoto (WKY) strain, has blood pressure that is similar to that of WKY. In the present studies we have investigated the role of angiotensin II and angiotensin-converting enzyme in the abnormal regulation of blood pressure in SHR-1DM. Systolic blood pressure was significantly elevated in SHR-1DM compared to SHR-1 and the mean blood pressure of SHR-1DM was reduced by enalapril (10 mg/kg per day for 14 days) to levels similar to those of SHR-1. Blood pressure was also significantly elevated in SHR-1DM compared to SHR-2. In SHR-1DM, basal plasma renin activity (PRA) and angiotensin II were similar to those of SHR-1, however, plasma angiotensin-converting enzyme (ACE) activity was significantly higher than in SHR-1. Enalapril increased PRA and angiotensin II and decreased ACE activity in SHR-1DM. The blood pressure-lowering effect of enalapril in SHR-1DM was significantly greater than in SHR-1 and the dose-response relationship for blood pressure to enalapril was similar in SHR-1DM and SHR-1.
